  10.  AB    A direct ELISA version of BioChem ImmunoSystems' synthetic peptide EIA
  11.        was evaluated to establish if detecting all immunoglobulin classes
  12.        improves the sensitivity of this test. HRP conjugates of 3 peptides from
  13.        HIV-1 GP120, HIV-1 GP41 and HIV-2 GP36 were used to detect antibodies
  14.        captured by these peptides on microtiter plates. Pending confirmatory
  15.        testing, the specificity of this EIA was 99.5% (1974 fresh samples
  16.        tested) while the sensitivity was 100% for HIV-1 (50/50) and 100% for
  17.        HIV-2 (15/15). By testing serially diluted samples the sensitivity of
  18.        the direct EIA was found to surpass that of the indirect EIA (version 1)
  19.        with seroconversion samples and was of similar sensitivity with samples
  20.        from later stages of HIV infection. The signal of both the direct and
  21.        indirect EIAs was reduced by > 90% after IgG absorption by Protein G
  22.        except with seroconversion samples where only a 30% signal reduction
  23.        occurred with the direct EIA. This indicated that the improved
  24.        sensitivity of the direct EIA with samples taken during seroconversion
  25.        is because the direct EIA format detects IgG as well as other
  26.        immunoglobulin classes.
