AB Distribution of nuclear ploidy in female mouse hepatocytes was
measured cytofluorimetrically using ethidium-bromide-stained
hepatocyte nuclei liberated by in situ collagenase perfusion of the
liver via the portal vein. After i.v. administration of lead
acetate or an i.p. general anaesthetic *(Valium* and Hypnorm), rapid
shifts of 8N and 4N nuclei to lower ploidy levels were recorded. It
was possible to block the ploidy changes with i.p. colchicine,
although no blocked metaphases were observed histologically. These
observations were consistent with an earlier report on changes in
nuclear ploidy induced by carbon tetrachloride (Steele et al.,
1981b). It is speculated that these changes in ploidy may represent
an early response of the hepatocytes to stimulation--an aspect of
liver behaviour of which we were not previously aware. Author.

AB This clinical study was performed on 13 neurosurgical patients. The
mean age was 56 years (range 45-70 years). After induction of
anesthesia the heart rate diminished by 21.4%, the mean pressure by
12.2% in the same time decreased the cardiac output by 19.3%, the
calculated total peripheral resistance increased by 13%. The oxygen
uptake decreased by 16%. The advantages and contraindications of
this technique in neuroanesthesia are discussed. Author.

AB In a controlled double-blind randomized study Thalamonal and *Valium*
were compared in 498 esophago-gastro-duodenoscopies. Both drugs
were injected intravenously immediately before the procedure.
Satisfactory sedation which made additional injection of the drug
during the endoscopy superfluous could be achieved in 66% of the
patients with Thalamonal and in 52% with *Valium* respectively. The
difference between the two preparations is statistically significant
(p less than 0,005). In 95 patients catamnestic studies were
performed in order to evaluate second episodes of fatigue after the
endoscopy. A good effect of the preparation during endoscopy and a
lack of second attacks of sedation following discharge was observed
only in 23% of the patients after Thalamonal and in 21% of the
patients after *Valium* injection, respectively. Therfore a good
premedication was achieved in only one fifth of the patients
receiving one of the two drugs. Author.

AB In 30 patients with congenital or acquired heart disease the
haemodynamic effects of diazepam *(Valium)* 0.3 mg/kg were
investigated during surgical procedures under neuroleptanalgesia.
The following parameters were measured or calculated: Heart rate
(HR), arterial pressure (-Part, Psyst, Pdiast), pulmonary artery
pressure (-PAP), right (-PRA) and left atrial pressure (-PLA), left
ventricular pressure (PLV), left ventricular enddiastolic pressure
(PLVED), left ventricular peak dp/dt (dp/dtmax), cardiac output
(CO), cardiac index (CI), stroke volume (SV), stroke index (SI),
total systemic resistance (TSR), total pulmonary resistance (TPR),
work index of the right (RVWI) and left ventricle (LVWI). In
comparison with a control group (n = 36) diazepam caused a decrease
in arterial pressure cardiac index, stroke index, right and left
atrial pressure and dp/dtmax. This, however, was mainly
attributable to vasodilatation and not to a negative inotropic
effect, which is of only minor importance with diazepam. These
haemodynamic changes resulted in a reduction in myocardial oxygen
consumption. Diazepam is a valuable drug in neuroleptanalgesia,
when an increase in blood pressure can not be controlled by *fentanyl*
or droperidol. Author.

AB A special technique of neurolept analgesia for electrocoagulation of
the gasserian ganglion is described which has been used since 1974.
Induction is by means of *Fentanyl* and *Valium.* The dosis is
sufficiently low for the patient to remain responsive and
co-operative but practically painfree. The actual thermocoagulation
is performed in intravenous methohexital sodium anaesthesia. The
dosis is kept so low that the patient wakes up within 2--3 minutes
and is able to inform the surgeon of the success or failure of the
operation. If no relief from pain has been obtained a further 20 mg
of methohexital are injected and the operation is repeated. Author.

AB The combination of *fentanyl* citrate (Sublimaze) and diazepam
*(Valium)* was evaluated for efficacy of analgesia, sedation, and
safety in 1,008 predominately outpatient urologic procedures. These
procedures included prostate biopsies, basket extractions of
ureteral calculi, internal urethrotomies, and cystourethroscopies.
Ninety-two per cent were judged to be successful with regard to
adequate tranquilization and relief of pain. No detrimental effects
were seen with the recommmended method and dosage. This drug
combination provides the clinician with an effective and safe
alternative to local, general, or spinal anesthesia for many routine
urologic procedures and allows them to becom true office procedures.
