- EMPLOYMENT RECORD

- September 1990 to current date:

- Plant Science Laboratory, Agronomy Department, University of Florida, Gaines-
- ville, Florida 32611. Retained as a Biological Scientist. My principal respon-
- sibility has been to develop a reliable Random Amplified Polymorphic DNA (RAPD)
- system. This system involves using the Polymerase Chain Reaction (PCR) and ran-
- dom 10 base primers to synthesize random DNA fragments from genomic DNA. The
- DNA fragments are subsequently separated by agarose gel electrophoresis and
- polyacrylamide Denaturing Gradient Gel Electrophoresis (DGGE). RAPD technology
- is used to distinguish intraspecific and interspecific differences in DNA and
- to follow the inheritance of DNA fragments. I have also been given the respon-
- sibility of identifying the presence of a desirable glutenin gene inherited
- from wheat in a wheat-rye cross. Identification of this glutenin gene involves
- the use of PCR and a set of primers specific for the gene. This system gener-
- ates a set of specific DNA fragments which are restricted to confirm the pre-
- sence of the glutenin gene. Other responsibilities include the extraction and
- purification of genomic DNA, digesting the DNA with restriction endonucleases,
- separating DNA fragments with agarose gel electrophoresis, blotting DNA onto
- nylon membranes, preparing and using radioactive probes for detecting sequences
- on the blots (RFLP analysis). Additional responsibilities include preparing
- genomic DNA and cDNA libraries, cloning specific DNA fragments, preparing DNA
- fragments for sequencing as well as using and maintaining a variety of labora-
- tory instruments. Finally, I am experienced in the use of IBM and VAX mainframe
- computers as well as microcomputers in data analysis.

- Duties involved quantifying the expression of enzymes of the photosynthetic

- rose synthesis pathways in extractions obtained from the leaves of plants
- exposed to varying CO2 concentrations and light intensities. Other responsibil-
- ities included teaching the laboratory sections of Integrated Principles of
- Biology BSC 2010C, Introductory Plant Physiology BOT 3503L and Plant Anatomy
- BOT 5225C.

- PERSONAL DATA

- I was born January 21, 1957 at Tampa, Florida and married September 14, 1979
- to Lisa M. Patterson. We have three children: Thomas J. Schweder, 12; Claira
- D. Schweder, 10 and; Alexandria P.A. Schweder, 8. We have lived in Gainesville,
- Florida since July 1987 so that I could attend graduate school at the Univer-
- sity of Florida. Lisa is a receiving clerk at the local Wal-Mart. My interests
- include computers, biking, swimming, SCUBA diving, sailing and photography.
