- FIGURE 6-5. Model of role of pRb in controlling G(1) to S transition.
- pRb is bound to both E2F and c-abl in the early G(1) phase of the
- cycle. Phosphorylation of pRb by activated cyclin/cdk complexes leads
- to release of E2F and c-abl. Release of the transcription factor, E2F,
- leads to transcriptional activation of genes required for transition
- from the G(1) phase of the cycle to S-phase. A role for c-abl in this
- transition has not yet been elucidated. There is a family of E2F
- proteins, numbered E2F-1 through 5, which can activate transcription
- when complexed with proteins of the DP family (DP-1,2, or 3).
- Potential target specificities for the different E2F/DP complexes are
- being investigated. Other proteins similar to pRb, such as p107 and
- p130, also bind to certain E2F factors, but do not bind to c-abl.
- There appears to be specificity for the different pRb-like proteins in
- terms of which E2F protein complexes are bound. These proteins may
- also release E2Fs at this stage in the cycle and thus participate in
- controlling the transition from G(1) to S.
