- FIGURE 13-8. PCR amplification of microsatellite repeats from the DNA
- of renal rhabdoid tumors and adjacent normal tissue. N marks the lanes
- obtained from normal tissue; T indicates the lanes obtained from
- tumor. (A) Loss of heterozygosity in the region of chromosome 22 band
- q11. Each allele characteristically shows a short ladder of bands
- probably generated as a result of slippage by DNA polymerase on the
- repeated sequences during amplification. The tumor shows apparent
- deletion of the allele responsible for the lower set of bands. (B)
- Microsatellite instability. The complex set of bands obtained from
- normal tissue probably arises from two alleles, a and b, which differ
- slightly in the number of repeats they contain. In the tumor, allele b
- has most likely expanded by several repeats to a size indicated by b'.
