- FIGURE 1-9. RNA fingerprinting and differential display. C127 mouse
- fibroblast cells stably transfected with mouse Met were stimulated
- with HGF/SF for 0, 1, 3, 8, or 24 hours (lanes 1-5). RNA was isolated
- and subjected to RT-PCR and RNA fingerprinting using a set of random
- primers. The resulting fragments were resolved by denaturing PAGE to
- create the fingerprints in each lane. Comparison of abundance of bands
- in the fingerprints shows that while the abundance of many RNAs is
- unchanged, some fluctuate (are differentially displayed) over the
- course of HGF/SF treatment. Cloning and sequencing of the DNAs present
- in the bands indicated with arrowheads reveals that these bands are
- two fragments of the IP10 chemokine gene. The expression pattern of
- IP10 over the course of HGF/SF stimulation was subsequently verified
