- FIGURE 1-4. (A) Steps in Southern analysis. 1. and 2.
- Restriction-enzyme-digested DNA samples are size fractionated by
- agarose gel electrophoresis. 3. DNA in the gel is denatured and then
- transferred from the gel to a membrane by capillary action. DNA is
- fixed to the membrane following transfer. 4. The membrane is
- hybridized with labeled probe, then washed to remove nonspecifically
- bound probe. 5. The washed and dried membrane is exposed to x-ray film
- for autoradiography. (B) Southern analysis detection of the mos
- oncogene in mouse, quail, and human cellular genomic DNA. Twenty
- micrograms each of mouse DNA digested with EcoRI, human DNA digested
- with HindIII, and quail DNA digested with BamHI were analyzed using
- the v-mos oncogene as a probe. The bands visible in each lane
- correspond to the fragment size which contains the corresponding c-mos
- genes.
