  1. FIGURE  1-10. (A)  RFLPs. A  diagram of human  chromosome 7  shows the
  2. position of the  met protooncogene at 7q31.  The sequence TCGA  is the
  3. recognition sequence of the TaqI  restriction enzyme. TaqI generates a
  4. 7.5-kb fragment in allele 1 and, due to  an internal site in allele 2,
  5. a 4- and 3.5-kb fragment in  allele 2. The probe is positioned  within
  6. the 4-kb fragment and detects the 7.5- and 4-kb fragments. (B)  Use of
  7. an RFLP in genetic diagnosis. The met gene is tightly linked to the CF
  8. gene; they  are therefore  almost always  inherited together.  In this
  9. family, allele  1 is on  the CF  chromosome and the  affected children
  10. (filled symbols) have a 1/1  genotype. Individuals with a 1/2 genotype
  11. are unaffected carriers and 2/2 children are unaffected noncarriers.
